چکیده

Bioassay for carcinogenic activity of a number of epoxides, lactones, hydroperoxides, and peroxides was reported from the laboratory at New York University (2,3). The compounds were tested by skin application on mice, s.c. injection in mice and rats, and feeding and intratracheal instillation in rats. Another series of related compounds was more recently tested at Fels Research Institute by s.c. injection in mice (1). Included in this series were compounds such as stearic acid and 7-stearolactone which, on the basis of known structurebiological activity relationships and the known occurrence Five compounds were selected: stearic acid, methyl stéarate, 7-stearolactone, p-nitroperbenzoic acid, and glycidyl stéarate. The same chemical samples and the same batch of tricaprylin as vehicle were used in both laboratories. The chemicals were given s.c. to mice once weekly for 26 weeks. The conditions of the experiments, as described in the full reports (1,3) were as identical as possible. The only known difference was that the New York University experiments were with ICR/Ha Swiss Millerton female mice, whereas the Fels experiments used CFW (Swiss Webster) female mice.

Workshop 4 . 3 Application of rat medium - term bioassays for detecting carcinogenic and modifying potentials of endocrine active substances *

Two in vivo bioassay methods, a rat medium-term liver bioassay and a rat multiorgan bioassay, can be used for detecting carcinogenic or modifying potentials of endocrine active substances (EASs) on endocrine disruption (ED). The first bioassay, the rat mediumterm liver bioassay, is fundamentally based on the two-step hypothesis of liver carcinogenesis; initiation with diethylnitrosamine (DEN, 2...

Carcinogenicity of oil shale tars, some of their components, and commercial products.

Bioassays for carcinogenicity of various primary processing products (crude oils or tars) and commercial products obtained from Estorian oil shale have been carried out since 1951. The products (undiluted or diluted) were painted twice weekly 50 times on the interscapular area of the skin of random-bred or CC57Br mice. The products processed at high temperatures have a higher carcinogenic activ...

Chronological supplement to the Carcinogenic Potency Database: standardized results of animal bioassays published through December 1982.

This paper is a chronological supplement to our earlier publication, "A Carcinogenic Potency Database of the Standardized Results of Animal Bioassays." We report here results of carcinogenesis bioassays published in Technical Reports of the National Cancer Institute/National Toxicology Program between July 1980 and December 1982, and the general literature between July 1981 and December 1982. Apparent correlation between structure and carcinogenicity of phenylenediamines and related compounds.

The carcinogenicity of 23 phenylenediamines and related compounds was reviewed. An extensive analysis of the methods used indicated that the bioassays were conducted well. The data suggest that the carcinogenicity of 4-substituted 1,3-phenylenediamines is reduced substantially or eliminated completely by oxidation of one or both amine groups or by N-substitution. Oxidation of a methyl substitue...

Application of genomic biomarkers to predict increased lung tumor incidence in 2-year rodent cancer bioassays.

Rodent cancer bioassays are part of a legacy of safety testing that has not changed significantly over the past 30 years. The bioassays are expensive, time consuming, and use hundreds of animals. Fewer than 1500 chemicals have been tested in a rodent cancer bioassay compared to the thousands of environmental and industrial chemicals that remain untested for carcinogenic activity. In this study,...
